A commonly used key figure for quality of dNTPs is "PURITY >99%" which states that all non-desired UV-absorbing species combined (such as other dNTPs, NTPs, or their mono-, di-, tetra- and polyphosphates) are present at levels below 1%.
Reporting reliable values of >99% for a dNTP lot however, requires sophisticated instrumentation and thorough protocols for analysis. At LAROVA a separate HPLC system is dedicated solely for detection of these impurities, and each dNTP lot is routinely analyzed by a sequence of programs that are optimized for high resolution of nucleosidic species.
A typical example is shown in the figure above. We analyzed a dATP lot of one of our competitors that was specified ">99%". While a standard analysis yields an apparent purity of 99.1%, the detected signals already suggest that resolution is incomplete (A). Re-analysis of the same dATP with one of our additional routine assays improves the resolution and the contaminants become visible and quantifiable and reveal that purity is clearly below specifications (B).
At Larova, only dNTPs that meet the 99% threshold in ALL QC assays are certified.
